Abstract:
(EN) The present invention provides a method of substantially retaining or otherwise preserving the biological activity of a dsRNA, present in a cell lysate, to post-transcriptionally silence the expression of a gene in a target organism, comprising the step of adding to the lysate a compound having the function of a protein – or amine - cross linking agent. The invention also comprises compositions comprising the lysate comprising dsRNA, and protein cross linking agents, as well as the use of said agents in the method.
(FR) La présente invention concerne un procédé permettant de retenir ou sinon préserver sensiblement l'activité biologique d'un ARNdb, présent dans un lysat cellulaire, pour inactiver de manière post-transcriptionnelle l'expression d'un gène dans un organisme cible, et comprenant l'étape consistant à ajouter au lysat un composé ayant la fonction d'un agent de réticulation de protéine ou amine. L'invention concerne également des compositions comprenant le lysat incluant l'ARNdb, et des agents de réticulation de protéines, ainsi que l'utilisation desdits agents dans le procédé.
