Abstract:

(EN)Provided herein are multispecific antigen-binding polypeptide constructs comprising at least two different heterodimers, each comprising a heavy chain and a light chain. At least one heterodimer comprises a Fab region comprising a lambda light chain and at least one heterodimer comprises a Fab region comprising a kappa light chain. One or more of the immunoglobulin heavy and light chains that form the antigen-binding polypeptide construct comprise amino acid modifications that promote correct pairing between the heavy and light chains to form the desired multispecific antigen-binding polypeptide construct. The amino acid modifications may be in the C_H1 and/or C_L domains, in the V_H and/or V_L domains, or a combination thereof.
(FR)L’invention concerne des constructions de polypeptides de liaison à l’antigène à spécificités multiples comprenant au moins deux hétérodimères différents, chacun comprenant une chaîne lourde et une chaîne légère. Au moins un hétérodimère comprend une région Fab comportant une chaîne légère lambda et au moins un hétérodimère comprend une région Fab comportant une chaîne légère kappa. Une ou plusieurs des chaînes légères et lourdes d’immunoglobuline qui forment la construction de polypeptide de liaison à l’antigène comprennent des modifications d’acides aminés qui favorisent l’appariement correct entre les chaînes lourdes et légères pour former la construction de polypeptide de liaison à l’antigène à spécificités multiples souhaitée. Les modifications d’acides aminés peuvent avoir lieu dans les domaines C_H1 et/ou C_L, dans les domaines V_H et/ou V_L, ou une de leurs combinaisons.
