Abstract:
(EN) The present invention relates to novel methods of diagnosing, preventing, and treating diseases, disorders, and infections relating to T-cell response. The disclosed methods for predicting MHC class II epitopes that elicit CD4+ T-cell response are based on the three-dimensional protein structure of an antigen of interest. Given such an antigen, structural properties of the protein taken from experimental and modeling data are used to compute an epitope likelihood score that characterizes the location of epitopes likely to elicit an immune response to CD4+ T-cells. The epitopes are then used to construct biomolecules, including peptides, which may be used to diagnose, prevent, and/or treat a number of diseases, disorders, and infections.
(FR) La présente invention concerne de nouveaux procédés pour diagnostiquer, prévenir et traiter des maladies, des troubles et des infections associés à une réponse de lymphocyte T Les procédés divulgués pour prédire des épitopes de classe II de MHC qui provoquent une réponse de lymphocyte T CD4+ sont fondés sur la structure protéique tridimensionnelle d’un antigène d’intérêt. Étant donné un tel antigène, des propriétés structurales de la protéine prises parmi des données expérimentales et de modélisation sont utilisées pour calculer un score de probabilité d’épitope qui caractérise l’emplacement d’épitopes susceptibles de provoquer une réponse immunitaire au niveau des lymphocytes T CD4+. Les épitopes sont ensuite utilisés pour construire des biomolécules, comprenant des peptides, qui peuvent être utilisées pour diagnostiquer, prévenir et/ou traiter un certain nombre de maladies, de troubles et d'infections.
