Abstract:
(EN) The present invention provides a method for in-vitro culturing and expanding natural killer (NK) cells in a cell culture medium comprising a population of NK cells, the method comprising a) adding an effective concentration of interleukin-21 (IL-21) at the beginning of the culturing process to said medium, b) adding repeatedly an effective concentration of interleukin-2 (IL-2) and/or interleukin-15 (IL-15) to said medium, and c) adding repeatedly feeder cells or membrane particles thereof to said medium, wherein said feeder cells are B cell derived which are EBV immortalized; and wherein said expansion of NK cells in said cell culture medium is maintained for at least 3 weeks.
(FR) La présente invention décrit un procédé de culture in vitro et d’expansion de cellules tueuses naturelles (NK) dans un milieu de culture de cellules comprenant une population de cellules NK, le procédé comprenant a) l’addition d’une concentration efficace d’interleukine-21 (IL-21) au début du processus de culture audit milieu, b) l’addition de manière répétée d’une concentration efficace d’interleukine-2 (IL-2) et/ou d’interleukine-15 (IL-15) audit milieu, et c) l’addition de manière répétée de cellules nourricières ou de particules membranaires de ces dernières audit milieu, lesdites cellules nourricières étant des cellules B dérivées qui sont immortalisées grâce à l’EBV ; et ladite expansion des cellules NK dans ledit milieu de culture de cellules est maintenue durant au moins 3 semaines.
