Title: LIBRARY CONSTRUCTION METHOD VIA BISULFITE SEQUENCING FOR RNA 5MC AND APPLICATION THEREOF
Abstract:
The present invention provides a library construction method via bisulfite sequencing for 5-methylcytosine (5mC) modification of an RNA molecule, a sequencing method and a methylation detection method. By using a random hexamer primer containing only three bases, A, C and T, the present invention significantly improves the reverse transcription and PCR amplification efficiency of a bisulfite-treated RNA fragment and the 5mC site detection efficiency.
