Abstract:
(EN) This disclosure relates to a chimeric antigen receptor for binding with a target antigen. The chimeric antigen receptor comprises at least one antigen specific targeting region including a multispecific antibody evolved from a wild-type antibody or a fragment thereof and having at least one of: (a) a decrease in activity in the assay at the normal physiological condition compared to the wild-type antibody or the fragment thereof, and (b) an increase in activity in the assay under the aberrant condition compared to the wild-type antibody or the fragment thereof. A method for using the chimeric antigen receptor and cytotoxic cells for cancer treatment is also provided.
(FR) La présente invention concerne un récepteur d'antigène chimérique destiné à se lier à un antigène cible. Le récepteur d'antigène chimérique comprend au moins une région de ciblage spécifique d'un antigène comprenant un anticorps multispécifique issu d'un anticorps de type sauvage ou d'un fragment de celui-ci et présentant au moins un effet parmi : (a) une diminution de l'activité dans le test dans des conditions physiologiques normales, par rapport à celle de l'anticorps de type sauvage ou d'un fragment de celui-ci, et (b) une augmentation de l'activité dans le test dans des conditions aberrantes, par rapport à celle de l'anticorps de type sauvage ou d'un fragment de celui-ci. L'invention concerne également un procédé d'utilisation du récepteur d'antigène chimérique et de cellules cytotoxiques pour le traitement du cancer.
