Abstract:
(EN) A multi-protein quantitation method based on equiponderous dimethylation marking. By means of a property that a dimethylation reaction has different rates on an amino group at a peptide N-terminal and an amino group on a lysine side chain at a peptide C-terminal, dimethylation marking of the peptide N-terminal and the C-terminal in an acid condition and an alkaline condition is implemented one after another; and multiple marking of a peptide sample is implemented by means of organic combination of a variety isotope forms of a dimethylation marking reagent. First-order spectrum mass-to-charge ratios of the peptide after multiple marking are completely the same, the mass-to-charge ratios of second-order spectrum fragment ions are different, and multiple quantitative analysis is carried out by extracting the corresponding second-order spectrum fragment ions. The method has the advantages: the quantitative accuracy is high, the precision is good, the dynamic range is wide, and simultaneous quantitative analysis of six protein samples can be simultaneously implemented, thereby greatly improving the flux of the quantitative analysis of protein, and saving analysis time.
(FR) L'invention concerne un procédé de quantification de protéines multiples basé sur un marquage de diméthylation à équi-pondération. Au moyen d'une propriété selon laquelle une réaction de diméthylation a des vitesses différentes sur un groupe amino à une extrémité N-terminale de peptide et un groupe amino sur une chaîne latérale de lysine à une extrémité C-terminale de peptide, des marquages de diméthylation de l'extrémité N-terminale et de l'extrémité C-terminale de peptide dans un état acide et un état alcalin sont mis en œuvre l'un après l'autre ; et un marquage multiple d'un échantillon de peptide est mis en œuvre au moyen d'une combinaison organique d'une variété de formes isotopes d'un réactif de marquage de diméthylation. Des rapports masse sur charge de spectre de premier ordre du peptide après un marquage multiple sont complètement identiques, les rapports masse sur charge d'ions fragmentés du spectre de second ordre sont différents, et une analyse quantitative multiple est effectuée par extraction des ions fragmentés du spectre de second ordre correspondant. Le procédé a les avantages selon lesquels : l'exactitude quantitative est élevée, la précision est bonne, la plage dynamique est grande, et l'analyse quantitative simultanée de six échantillons de protéines peut être mise en œuvre de manière simultanée, ce qui permet d'améliorer considérablement le flux de l'analyse quantitative de protéines, et d'économiser du temps d'analyse.
(ZH) 一种基于等重二甲基化标记的多重蛋白质定量方法,利用酸性条件下二甲基化反应在肽段N末端氨基和肽段C末端赖氨酸侧链氨基上速率不同的性质,在酸、碱条件下先后实现肽段N末端和C末端的二甲基化标记,通过二甲基化标记试剂各种同位素形式有机组合实现肽段样品的多重标记。多重标记后的肽段一级谱质荷比完全相同,二级谱碎片离子的质荷比存在差异,提取相应二级谱碎片离子强度进行多重定量分析。该方法的优势在于:定量准确度高,精密度好,动态范围宽,可以同时实现六重蛋白质样品的同时定量分析,大大提高蛋白质定量分析的通量,节约分析时间。
