Abstract:
(EN) Disclosed is the use of the interaction between the influenza virus proteins and the host protein CPSF30 in the inhibition of the tumor cells proliferation. The two viral proteins NS1 and NP of influenza virus inhibit the normal activity of the host protein CPSF30 by binding to CPSF30, thereby blocking the cell cycle of the host cell at the G0/G1 phase, and further inhibiting the division of the host cell. By expressing exogenous NS1 and NP proteins in cancer cell lines or by inhibiting of the expression of endogenous CPSF30 with siRNA, the cell cycle can be arrested at G0/G1 phase and the proliferation of cancer cells can be slowed.
(FR) La présente invention concerne l'utilisation de l'interaction entre les protéines du virus de la grippe et la protéine hôte CPSF30 dans l'inhibition de la prolifération de cellules tumorales. Les deux protéines virales NS1 et NP du virus de la grippe inhibent l'activité normale de la protéine hôte CPSF30 par liaison à CPSF30, ce qui permet de bloquer le cycle cellulaire de la cellule hôte au niveau de la phase G0/G1, et d’inhiber en outre la division de la cellule hôte. Par l’expression de protéines NS1 et NP exogènes dans des lignées cellulaires cancéreuses ou par l’inhibition de l'expression de CPSF30 endogène par de petits ARNi, le cycle cellulaire peut être arrêté au niveau de la phase G0/G1 et la prolifération de cellules cancéreuses peut être ralentie.
(ZH) 本发明公开了流感病毒蛋白与宿主蛋白CPSF30的相互作用在抑制癌细胞增殖中的应用。流感病毒的NS1和NP两个病毒蛋白通过与宿主蛋白CPSF30结合,抑制CPSF30的正常活性,从而将宿主细胞的细胞周期抑制在G0/G1期,进而抑制宿主细胞的分裂。通过在癌细胞系中表达外源的NS1和NP蛋白,或者将细胞内源性的CPSF30通过siRNA的方法进行表达抑制,可以将细胞周期停滞在G0/G1期并减缓癌细胞的增殖。
