Abstract:
(EN) The invention discloses diagnostic techniques based on single cell genomics, consisting of obtaining a blood sample, enriching a sub-population of cells present in the blood sample, sequestering individual cells or group of cells from the blood sample, obtaining sequencing data from the sequestered cells or group of cells, using genetic variant information to determine the provenance of the cells, and genetically analyzing the cells of the correct provenance to provide a diagnostic readout. Using the cell-based testing techniques of the invention, the number of false positives is greatly reduced when compared to cell-free DNA (cfDNA) based traditional testing techniques. The invention may be effectively employed for non-invasive prenatal (NIPT) diagnostics, oncological testing and other diagnostic procedures.
(FR) L'invention concerne des techniques de diagnostic fondées sur la génomique de cellule unique, constituées par l'obtention d'un échantillon de sang, l'enrichissement d'une sous-population de cellules présentes dans l'échantillon de sang, la séquestration des cellules individuelles ou d'un groupe de cellules à partir de l'échantillon de sang, l'obtention de données de séquençage à partir des cellules ou du groupe de cellules séquestré(es), à l'aide des informations de variants génétiques afin de déterminer la provenance des cellules, et l'analyse génétique des cellules de la bonne provenance pour fournir une lecture de diagnostic. En mettant en œuvre les techniques d'essai à base de cellules de l'invention, le nombre de faux positifs est considérablement réduit par rapport aux techniques d'essai classiques fondées sur l'ADN acellulaire (ADNac). L'invention peut être utilisée efficacement pour des diagnostics prénataux non invasifs (NIPT), un essai oncologique et d'autres procédures de diagnostic.
