Abstract

A candida parapsilosis CGMCC 9630. The bacterial strain expresses a carbonyl reductase and a coding gene and amino acid sequence of the carbonyl reductase, comprises a recombinant expression vector and a recombinant expression conversion medium of a gene sequence, and utilizes a whole cell of the candida parapsilosis, the carbonyl reductase thereof or the corresponding recombinant conversion medium to serve as a catalyst to catalyze an asymmetric reduction of a prochiral carbonyl compound, especially for reduction of 6-carbonyl-8-halogenated octanoate to prepare (R)-α-thioctic acid and to synthesize a precursor (R)-6-carbonyl-8-halogenated octanoate. Compared to other methods for preparing the (R)-6-carbonyl-8-halogenated octanoate via asymmetric reduction, the method has a high substrate concentration, a mild reaction condition, a high productivity and a high product optical purity, is environmentally friendly, and has a good application prospect in industrial production of (R)-α-thioctic acid.

L'invention concerne un Candida parapsilosis CGMCC 9630. La souche bactérienne exprime une carbonyl réductase et un gène codant et une séquence d'acides aminés de la carbonyl réductase, comprend un vecteur d'expression recombinant et un milieu de conversion d'expression recombinant d'une séquence génique et utilise une cellule entière de candida parapsilosis, la carbonyl réductase correspondante ou le milieu de conversion recombinant correspondant pour servir en tant que catalyseur pour catalyser une réduction asymétrique d'un composé carbonyle prochiral, en particulier pour la réduction d'octanoate 6-carbonyl-8-halogéné pour préparer de l'acide (R)-α-thioctique et pour synthétiser un précurseur d'octanoate (R)-6-carbonyl-8-halogéné. Par rapport à d'autres procédés de préparation de l'octanoate (R)-6-carbonyl-8-halogéné par réduction asymétrique, le procédé présente une concentration élevée en substrat, une condition de réaction douce, une productivité élevée et une haute pureté optique, est respectueux de l'environnement et possède une bonne perspective d'application dans la production industrielle de l'acide (R)-α-thioctique.

近平滑假丝酵母CGMCC 9630,该菌株表达羰基还原酶及其编码基因和氨基酸序列,含有该基因序列的重组表达载体和重组表达转化体,以及利用所述近平滑假丝酵母整细胞、其羰基还原酶或相应的重组转化体作为催化剂用于催化潜手性羰基化合物的不对称还原,特别是6-羰基-8-卤代辛酸酯还原制备(R)-α-硫辛酸合成前体(R)-6-羟基-8-卤代辛酸酯。与不对称还原制备(R)-6-羟基-8-卤代辛酸酯的其他方法相比,该方法具有底物浓度高、反应条件温和、环境友好、产率高、产品光学纯度高等优势,因此在(R)-α-硫辛酸的工业化生产中具有很好的应用前景。
