Methods, devices and systems for analyzing precious samples of cells, including single cells are provided. The methods, devices, and systems in various embodiments of the invention are used to assess genomic heterogeneity, which has been recognized as a central feature of many cancers and plays a critical role in disease initiation, progression, and response to treatment. The methods devices and systems are also used to analyze embryonic biopsies for reimplantation genetic diagnosis (PGD). In one embodiment, the devices, systems and methods provided herein allow for the construction of genomic and RNA-seq libraries without a pre- amplification step.

L'invention concerne des procédés, des dispositifs et des systèmes pour analyser des échantillons précieux de cellules, comprenant des cellules uniques. Les procédés, les dispositifs et les systèmes dans divers modes de réalisation de l'invention sont utilisés pour évaluer l'hétérogénéité génomique, qui a été reconnue en tant que caractéristique centrale de nombreux cancers et qui joue un rôle critique au début d'une maladie, sa progression et la réponse au traitement. Les procédés, dispositifs et systèmes sont également utilisés pour analyser des biopsies embryonnaires en vue d'un diagnostic génétique préimplantatoire (DPI). Dans un mode de réalisation, les dispositifs, systèmes et procédés de la présente invention permettent la construction de bibliothèques génomiques et de séquences d'ARN sans étape de préamplification.
