Abstract:
(EN) A method and system are provided for detecting the concentration of an anaiyte in a fluid sample. The method and system involve analysis of a volatilized, ionized fluid sample using a mass spectrometer or other ionic anaiyte detection device that provides a signal proportional in intensity to the quantity of ionized anaiyte detected. The improvement involves replacement of a necessary non-analyte component in the fluid sample with a substitute component that serves the same purpose as the original component but is either more volatile than the original component and/or the anaiyte or undergoes a reaction to provide lower molecular weight reaction products, and results in an increased intensity in signal and signal-to-noise ratio. Acoustic fluid ejection is a preferred method of generating nanoliter-sized droplets of fluid sample that are then volatilized, ionized, and analyzed.
(FR) L’invention concerne un procédé et un système de détection de la concentration d’un analyte dans un échantillon de fluide. Le procédé et le système impliquent l’analyse d’un échantillon de fluide ionisé et volatilisé au moyen d’un spectromètre de masse ou d’un autre dispositif de détection d’analyte ionique qui émet un signal dont l’intensité est proportionnelle à la quantité d’analyte ionisé détecté. L’amélioration implique le remplacement d’un composant nécessaire qui n’est pas l’analyte dans l’échantillon de fluide par un composant de substitution qui remplit le même rôle que le composant original mais qui est plus volatil que le composant original et/ou l’analyte ou qui est soumis à une réaction dont les produits de réaction présentent un poids moléculaire inférieur, et qui permet d’accroître l’intensité de signal et le ratio signal-bruit. L’éjection de fluide acoustique est un procédé préféré de génération de gouttelettes d’échantillon de fluide de l’ordre du nanolitre qui sont alors volatilisées, ionisées, et analysées.
