(EN)The present invention relates to: a vector expressing, on the surface of a microorganism, matrix 2 (M2) and hemagglutinin (HA2), which are antigen proteins specific to the influenza virus; and a microorganism transformed thereby and, more specifically, to: a surface expression vector comprising a poly-γ-glutamic acid synthetase complex gene (pgsA), a cholera toxin A1 subunit (CTA1), and genes encoding M2 and HA2, which are antigen proteins specific to the influenza virus; a recombinant microorganism transformed by the vector; and a vaccine for preventing an influenza virus infection, containing an antigen, which is obtained from the recombinant microorganism, as an active ingredient. According to the present invention, the microorganism in which M2 and HA2 (CTA1-sM2HA2) antigen proteins bound to CTA1 are surface-expressed causes an excellent immunity reaction in various influenza A virus subtypes, and a microorganism surface-expressing M2 and HA2, which are antigen proteins specific to the influenza virus, is useful as a mucosal vaccine capable of direct oral or nasal application.
(FR)La présente invention concerne : un vecteur exprimant, à la surface d'un micro-organisme, la matrice 2 (M2) et l'hémagglutinine (HA2), qui sont des protéines antigéniques spécifiques au virus de la grippe ; et un micro-organisme transformé par celui-ci et plus particulièrement : un vecteur d'expression de surface comprenant un gène du complexe d'acide poly-γ-glutamique synthétase (pgsA), une sous-unité A1 de la toxine cholérique (CTA1) et des gènes codant pour M2 et pour HA2, qui sont des protéines antigéniques spécifiques au virus de la grippe ; un micro-organisme recombinant transformé par le vecteur ; et un vaccin destiné à la prévention d'une infection par le virus de la grippe, contenant un antigène, qui est obtenu à partir du micro-organisme recombinant, en tant qu'ingrédient actif. Selon la présente invention, le micro-organisme, dans lequel les protéines antigéniques M2 et HA2 (CTA1-sM2HA2) liées à CTA1 sont exprimées en surface, provoque une excellente réaction d'immunité dans divers sous-types du virus de la grippe A et un micro-organisme exprimant en surface M2 et HA2, qui sont des protéines antigéniques spécifiques au virus de la grippe, est utile en tant que vaccin par voie muqueuse permettant une application buccale ou nasale directe.
(KO)본 발명은 인플루엔자 바이러스의 특이 항원 단백질인 매트릭스 2(M2) 및 헤마글루티닌(HA2)을 미생물의 표면에 발현시키는 벡터 및 이에 의해 형질전환된 미생물에 관한 것으로, 더욱 자세하게는 폴리감마글루탐산 합성효소 복합체 유전자(pgsA), 콜레라 독소 A1 서브유닛(CTA1), 인플루엔자 바이러스의 특이 항원 단백질인 매트릭스 2(M2) 및 헤마글루티닌(HA2)을 코딩하는 유전자를 포함하는 표면 발현벡터, 상기 벡터로 형질전환된 재조합 미생물 및 상기 재조합 미생물로부터 수득한 항원을 유효성분으로 함유하는 인플루엔자 바이러스 감염의 예방용 백신에 관한 것이다. 본 본 발명에 따른 콜레라 독소 A1 서브유닛과 결합된 매트릭스 2 및 헤마글루티닌(CTA1-sM2HA2) 항원단백질이 표면 발현된 미생물은 다양한 A형 인플루엔자 바이러스 서브타입에 우수한 면역반응을 유발하며, 상기 인플루엔자 바이러스의 특이 항원단백질인 매트릭스 2 및 헤마글루티닌을 표면 발현하는 미생물은 경구 또는 비강에 직접 적용할 수 있는 점막백신으로 유용하다.
