Abstract:
(EN)  The present invention provides a method for improving the efficiency of inducing a pluripotent stem cell, and a vector and composition used for the method. The inventors succeeded in significantly increasing the efficiency of inducing a pluripotent stem cell by further introducing a vector including a KLF gene and not including an OCT gene and a SOX gene in pluripotent stem cell induction including a step for introducing a vector including a KLF gene, an OCT gene, and a SOX gene in this order. This method is capable of efficiently inducing a pluripotent stem cell under temperature conditions closer to a physiological environment, and has excellent characteristics in that the vector is rapidly eliminated after pluripotent stem cell induction. The present invention makes it possible to induce a pluripotent stem cell with greater efficiency.
(FR) L'invention fournit un procédé améliorant le taux d'induction de cellules souches pluripotentes, et un vecteur ainsi qu'une composition mis en œuvre dans ce procédé. Lors de l'induction de cellules souches pluripotentes incluant une étape au cours de laquelle est induit un vecteur contenant dans l'ordre un gène KLF, un gène OCT et un gène SOX, un vecteur contenant un gène KLF mais exempt de gène OCT et de gène SOX, étant également introduit, le taux d'induction de cellules souches pluripotentes est élevé avec succès et de manière significative. Le procédé de l'invention est caractéristique en ce qu'il permet d'induire efficacement des cellules souches pluripotentes sous des conditions de températures plus proches d'un environnement physiologique, et qu'il est excellent en termes de retrait rapide de vecteur après induction des cellules souches pluripotentes. Selon l'invention, il est donc possible d'effectuer une induction de cellules souches pluripotentes de manière bien plus efficace.
(JA)  本発明は、多能性幹細胞の誘導効率を改善する方法およびそのために用いられるベクターおよび組成物を提供する。KLF遺伝子、OCT遺伝子、およびSOX遺伝子を、この順序で含むベクターを導入する工程を含む多能性幹細胞の誘導において、KLF遺伝子を含みOCT遺伝子およびSOX遺伝子を含まないベクターをさらに導入することにより、多能性幹細胞の誘導効率を有意に上昇させることに成功した。本発明の方法は、より生理環境に近い温度条件で効率よく多能性幹細胞を誘導することが可能で、多能性幹細胞を誘導後は、速やかにベクターが除去されるという優れた特徴を有する。本発明により、一段と効率的に多能性幹細胞の誘導を行うことが可能となった。
