Abstract:
(EN) A proteomic expression platform to identify age-related sepsis risk is disclosed using patients with community-acquired pneumonia. A semi-quantitative plasma proteomics workflow was applied which incorporated tandem immuno affinity depletion, iTRAQ labeling, strong cation exchange fractionation, and nanoflow-liquid chromatography coupled to high resolution mass spectrometry. A protein profile was determined that exhibit statistically significant differences in expression levels amongst patients with severe sepsis as a function of age. Representative pathways that are differentially-expressed include, but are not limited to, acute phase response, coagulation signaling, atherosclerosis signaling, lipid metabolism, and production of nitric oxide/reactive oxygen species.
(FR) La présente invention concerne une plate-forme d'expression protéomique pour identifier le risque septique lié à l'âge auprès de patients atteints de pneumopathies communautaires. Une analyse protéomique semi-quantitative du plasma a été réalisée, laquelle a incorporé la déplétion par immunoaffinité en tandem, le marquage iTRAQ, le fractionnement par échange de cations forts et la nano-chromatographie liquide couplée à une spectrométrie de masse haute résolution. Un profil protéique a été déterminé et présente des différences statistiquement significatives dans les niveaux d'expression en fonction de l'âge parmi les patients atteints de sepsis sévère. Les voies représentatives qui sont exprimées différentiellement comprennent, sans toutefois s'y limiter, la réponse de phase aiguë, la signalisation dans la coagulation, la signalisation dans l'athérosclérose, le métabolisme des lipides et la production d'espèces d'oxyde nitrique/oxygène réactif.
