Title: METHOD FOR SCREENING YEAST STRAIN HAVING IMPROVED RECOMBINANT PROTEIN SECRETION CAPACITY USING CELL WALL DEFICIENT YEAST MUTANT
Abstract:
The present invention relates to a method for screening a yeast strain secreting an introduced target protein, a method for improving the secretion capacity of a target protein, a method for producing a target protein, and a method for producing a yeast mutant having an improved capacity to secreting Gas1 or a fusion protein thereof. With respect to the yeast strain of the present invention, which expresses a fusion protein in which a target protein produced by using a Gas1-deficient strain of which the growth is inhibited under the cell wall stress condition is bound to the N-terminal of the Gas1 protein, the increase in the growth capacity of the yeast strain is verified by forming a cell wall due to the secretion of the fusion protein, thereby effectively verifying the yeast strain having an increased secretion capacity. Thus, the present invention can be useful in selecting strains having an improved secretion capacity from a series of variant libraries, which are prepared by random mutagenesis for mass-producing recombinant proteins.
