(EN)This invention relates to an improved method for manufacturing a matured protein with its intact sequence from a fusion protein. More specifically, the present invention relates to enzymatic methods using picornaviral 3C protease and Xaa-Pro dipeptidyl aminopeptidase for the removal of an N-terminal extension from a fusion protein, and the use of these methods to produce recombinant products with the correct N-terminal sequence.
(FR)L'invention concerne un procédé amélioré qui permet de fabriquer une protéine mature avec sa séquence intacte à partir d'une protéine de fusion. Plus particulièrement, la présente invention concerne des procédés enzymatiques qui font intervenir la protéase 3C picornavirale et la dipeptidyl-aminopeptidase Xaa-Pro pour l'élimination d'une extension N-terminale d'une protéine de fusion, et l'utilisation de ces procédés pour produire des produits de recombinaison avec la séquence N-terminale correcte.
