The invention is directed to methods, reagents, and kits for the detection of MAPK/ERK pathway mutations in a patient diagnosed for cancer. In one embodiment, the invention comprises a sensitive and selective method to identify mutations to the BRAF, KRAS, and NRAS genes in a single reaction. In another aspect of this embodiment, the invention comprises primers and probes for the detection of the BRAF, KRAS, and NRAS mutations using a single nucleotide primer extension assay. In another embodiment the invention is used to identify and select patients amenable for treatment with an ERK inhibitor.

L'invention porte sur des procédés, des réactifs et des trousses destinés à la détection de mutations de la voie MAPK/ERK chez un patient chez qui un cancer a été diagnostiqué. Dans un mode de réalisation, l'invention concerne un procédé sensible et sélectif qui permet d'identifier des mutations des gènes BRAF, KRAS et NRAS en une seule réaction. Selon un autre aspect de ce mode de réalisation, l'invention se rapporte à des amorces et à des sondes permettant de détecter des mutations de BRAF, KRAS et NRAS en un seul essai d'extension d'amorce nucléotidique. Dans un autre mode de réalisation, l'invention permet d'identifier et de sélectionner des patients pouvant faire l'objet d'un traitement par un inhibiteur de ERK.
