Abstract

Provided is an iPS precursor cell capable of highly efficient transgenesis as a result of stable maintenance cultivation. The precursor cell is prepared by introducing either OCT4, SOX2, or KLF4 to a human somatic cell, and culturing same in a medium including an MEK inhibitor, a GSK3β inhibitor, and a leukemia inhibitor factor (LIF). The MEK inhibitor is, for example, PD0325901. The GSK3β inhibitor is, for example, CHIR99021. The iPS precursor cell is OCT4 negative, NANOG negative, and NESTIN positive.

L'invention concerne une cellule précurseur d'IPS apte à une transgénèse hautement efficace comme résultat d'une culture d'entretien stable. La cellule précurseur est préparée par l'introduction soit d'OCT4, SOX2, soit KLF4 à une cellule somatique humaine, et la culture de celle-ci dans un milieu comprenant un inhibiteur de MEK, un inhibiteur de GSK3β et un facteur inhibiteur de la leucémie (LIF). L'inhibiteur de MEK est, par exemple, PD0325901. L'inhibiteur de GSK3β est, par exemple, CHIR99021. La cellule précurseur d'iPS est négative pour OCT4, négative pour NANOG et positive pour NESTINE.

 安定して維持培養できることにより、高効率な遺伝子導入が可能であるiPS前駆細胞を提供する。ヒト体細胞へOCT4、SOX2、及びKLF4のうち何れか一つを導入し、MEK阻害剤、GSK3β阻害剤、及び白血病阻害因子LIFを含む培地にて培養することにより作製される。MEK阻害剤は、例えばPD0325901である。GSK3β阻害剤は、例えばCHIR99021である。iPS前駆細胞は、OCT4陰性、NANOG陰性、NESTIN陽性である。
