Abstract

Methods for diagnosis, prevention, and treatment of narcolepsy are disclosed. In particular, the invention relates to methods of detecting T cells reactive to narcolepsy-inducing antigens, such as hypocretin or influenza H1 peptides, displayed in complexes with human leukocyte antigen DQ0602. The invention further relates to methods of treating subjects for narcolepsy by blocking formation of DQ0602-hypocretin and DQ0602-influenza H1 peptide epitope complexes and their interaction with their cognate T cell receptor (TCR).

La présente invention concerne des procédés de diagnostic et de prévention, et des méthodes de traitement, de la narcolepsie. En particulier, l'invention porte sur des méthodes de détection de lymphocytes T réactifs aux antigènes induisant la narcolepsie, tels que l'hypocrétine ou les peptides de la grippe de type H1, présents dans des complexes avec l'antigène leucocytaire humain DQ0602. L'invention porte en outre sur des méthodes de traitement de sujets atteints de narcolepsie par le blocage de la formation de la DQ0602-hypocrétine et des complexes DQ0602-épitope peptidique de la grippe de type H1, ainsi que de leur interaction avec leur récepteur connu de l'antigène des lymphocytes T (TCR).
