Abstract

EstATII is an esterase that a halotolerant, thermophilic and resistant to a spectrum of heavy metals including toxic concentration of metals. It was isolated from the lowest convective layer of the Atlantis II Red Sea brine pool. The Atlantis II brine pool is an extreme environment that possesses multiple harsh conditions such as; high temperature, salinity, pH and high concentration of metals, including toxic heavy metals. A fosmid metagenomic library using DNA isolated from the lowest convective layer this pool was used to identify EstATII. Polynucleotides encoding EstATII and similar esterases are disclosed and can be used to make EstATII. EstATII or compositions or apparatuses that contain it may be used in various processes employing lipases/esterases especially when these processes are performed under harsh conditions that inactivate other kinds of lipases or esterases.

La présente invention se rapporte à l'estérase EstATII qui est une estérase qui est halotolérante, thermophile et résistance au spectre des métaux lourds comportant une concentration toxique en métaux. Elle a été isolée de la couche convective la plus basse du bassin de saumure Atlantis II de la mer Rouge. Le bassin de saumure Atlantis II constitue un environnement extrême qui possède de multiples conditions sévères telles qu'une température élevée, un certain taux de salinité, un certain pH et une concentration élevée en métaux, y compris en métaux lourds toxiques. Une banque fosmidique métagénomique qui utilise l'ADN isolé de ce bassin de la couche convective la plus basse, a été utilisée pour identifier l'estérase EstATII. Des polynucléotides codant l'estérase EstATII et des estérases similaires sont divulgués et peuvent être utilisés pour réaliser l'estérase EstATII. L'estérase EstATII ou des compositions ou des appareils qui la contiennent peuvent être utilisés dans divers procédés qui utilisent les lipases/estérases, en particulier lorsque ces procédés sont effectués dans des conditions extrêmes qui inactivent les autres types de lipases ou d'estérases.
