(EN)The present invention provides nucleic acid constructs, expression vectors, transgenic cell and methods of making and using the same, wherein the nucleic acid construct includes a synthetic promoter designed using selected PDX-1 activation sites such as those observed in the human insulin promoter (HIP). In illustrative working embodiments of the invention, an exogenous nucleic acid fragment encoding thymidine kinase is operably linked to the synthetic promoter which is then shown to regulate the expression of this polypeptide.
(FR)La présente invention concerne des constructions d'acides nucléiques, des vecteurs d'expression, des cellules transgéniques et leurs procédés de fabrication et d'utilisation, ladite construction d'acide nucléique comprenant un promoteur synthétique conçu au moyen de sites d'activation de PDX-1 sélectionnés, tels que ceux observés dans le promoteur du gène humain de l'insuline (HIP). Dans des exemples de réalisations fonctionnelles de l'invention, un fragment d'acide nucléique exogène codant pour la thymidine kinase est en liaison fonctionnelle avec le promoteur synthétique dont on démontre qu'il régule l'expression dudit polypeptide.
