Abstract

A method to measure all relevant biologic hydrogen sulfide pools, namely free hydrogen sulfide, acid-labile sulfide, and bound sulfane sulfur, has been developed. This new method involves selective liberation, trapping and derivatization of labile hydrogen sulfide. The total labile sulfide, including the contribution of the bound sulfane sulfur pool, the acid-labile pool, and free H2S, was measured by incubating the sample with a reducing agent, TCEP (Tris(2-carboxyethyl)phosphine hydrochloride), to reduce disulfide bonds in an acid solution. This method was used to measure the three sulfide pools in blood samples from mice and from humans. This method can be used for research, environmental, and clinical diagnostic purposes in determining hydrogen sulfide bioavailability in biological or other samples.

Cette invention concerne un procédé permettant de mesurer tous les pools de sulfures d'hydrogène biologiques pertinents, à savoir sulfure d'hydrogène libre, sulfure labile en milieu acide, et sulfane-soufre lié. Ce nouveau procédé implique la libération sélective, le piégeage et la dérivatisation du sulfure d'hydrogène labile. Le sulfure labile total, comprenant la contribution du pool de sulfane-soufre lié, du pool labile en milieu acide, et du H2S libre, a été mesuré par incubation de l'échantillon avec un agent de réduction, le TCEP (chlorhydrate de tris(2-carboxyéthyl)phosphine), pour réduire les liaisons disulfure dans une solution acide. Ce procédé a été utilisé pour mesurer les trois pools de sulfures dans des échantillons de sang provenant de souris et de sujets humains. Il peut être utilisé dans le cadre de la recherche, et à des fins environnementales, et diagnostiques cliniques pour déterminer la biodisponibilité du sulfure d'hydrogène dans des échantillons biologiques ou autres.
