Abstract:
(EN) The invention relates to a calcium sensor based on a fusion protein which includes: (i) a first polypeptide domain which is capable of bonding Ca 2+, (ii) a second polypeptide domain of sequence SEQ ID NO. 1 (GFPuv) or a variant thereof that maintains two maximums in the excitation spectrum thereof and at least one maximum in the emission spectrum thereof, and (iii) a flexible peptide that bonds the first domain and the second domain in a covalent manner. The invention also provides a genetically modified non-human animal that includes the fusion protein. Likewise, the invention provides methods for detecting the concentration of intracellular calcium Ca 2+.
(FR) La présente invention concerne un capteur de calcium à base d'une protéine de fusion comprenant (i) un premier domaine polypeptidique présentant la capacité de lier Ca2+, (ii) un second domaine polypeptidique de séquence SEQ ID NO: 1 (GFPuv) ou une variante de celle-ci qui maintient deux maximums dans son spectre d'excitation et au moins un maximum dans son spectre d'émission, et (iii) un peptide flexible qui lie le premier domaine et le second domaine de manière covalente. L'invention concerne également un animal transgénique non humain qui comprend la protéine de fusion. Elle concerne aussi des procédés permettant de détecter la concentration en calcium Ca2+ intracellulaire.
(ES) La invención proporciona un sensor de calcio basado en una proteína de fusión que comprende (i) un primer dominio polipeptídico con capacidad de unir Ca 2+, (ii) un segundo dominio polipeptídico de la secuencia SEQ ID NO: 1 (GFPuv) o una variante de la misma que mantenga dos máximos en su espectro de excitación y al menos un máximo en su espectro de emisión, y (iii) un péptido flexible queune el primer dominio y el segundo dominio de manera covalente. También proporciona un animal no humano transgénico que comprende la proteína de fusión. Asimismo, la invención proporciona métodos para detectar la concentración de calcio Ca 2+ intracelular.
