Abstract:
(EN) Compositions and methods are provided for nucleotide analogs comprising protein shields for improving enzyme photostability in single molecule real time sequencing. Nucleotide analogs of the invention have a protein shield between the dye moieties and nucleotide moieties of the analog. The protein prevents the direct interaction of the dye moiety with the enzyme carrying out nucleotide synthesis preventing photodamage to the enzyme. The nucleotide analogs of the invention can have multiple dyes and multiple nucleotide moieties.
(FR) La présente invention concerne des compositions et des procédés destinés à des analogues nucléotidiques comprenant des boucliers protéiques permettant d'améliorer la photostabilité des enzymes lors du séquençage en temps réel d'une molécule individuelle. Les analogues nucléotidiques de l'invention possèdent un bouclier protéique situé entre les fragments de colorant et les fragments de nucléotide de l'analogue. La protéine permet d'éviter l'interaction directe du fragment de colorant avec l'enzyme effectuant la synthèse nucléotidique, ce qui permet d'éviter l'endommagement de l'enzyme par la lumière. Les analogues nucléotidiques de l'invention peuvent contenir plusieurs colorants et plusieurs fragments de nucléotide.
