Abstract:
(EN) The present invention provides methods for conducting screens using nucleic acid elements (e.g., interfering RNAs) to confidently identify hit genetic elements. The present invention further comprises constructing vectors that contain two or more nucleic acid elements to knock down all pairwise combinations of the hit genetic elements identified from the screen. Following quantitation of the single and double-knockdown phenotypes, genetic interactions between all gene pairs can be calculated. Genes can then be clustered according to the similarity of the pattern of their interactions with all of the other genes to obtain a genetic interaction map, which can advantageously be used to predict functional associations between genes and identify drug targets for therapy such as combination cancer therapy.
(FR) La présente invention concerne des procédés pour la réalisation de criblages à l'aide d'éléments acides nucléiques (par exemple des ARN interférents) pour identifier avec confiance des éléments génétiques atteints. La présente invention concerne en outre la construction de vecteurs qui contiennent au moins deux éléments acides nucléiques pour inactiver toutes les combinaisons appariées des éléments génétiques atteints identifiés à partir du crible. Suivant l'analyse quantitative des phénotypes à inactivation simple et double, des interactions génétiques entre toutes les paires de gènes peuvent être calculées. Des gènes peuvent être ensuite regroupés selon la similarité du motif de leurs interactions avec tous les autres gènes pour obtenir une carte d'interaction génétique, qui peut être utilisée de façon avantageuse pour prédire des associations fonctionnelles entre des gènes et identifier des cibles de médicament pour une thérapie, telle qu'une polythérapie anticancéreuse.
