Abstract:
(EN) A method of conjugating a substrate includes exchanging a counter ion associated with a biomolecule with a lipophilic counter ion to form a biomolecule complex, dispersing the biomolecule complex in a non¬ aqueous solvent, and coupling the biomolecule complex to a sub¬ strate in the presence of the nonaqueous solvent. The substrate or solid support is a polymeric particle, the biomolecule to be immo¬ bilized on the particle is an oligonucleotide, the lipophilic counter ion is a tetrabutylammonium ion and the nonaqueous solvent is N- methylpyrrolidone (NMP). The oligonucleotide coated particles are used in nucleic acid isolation or in nucleic acid sequencing reactions.
(FR) Un procédé de conjugaison d'un substrat de l'invention consiste à échanger une contre-ion associé à une biomolécule contre un contre-ion lipophile pour former un complexe de biomolécules, à disperser le complexe de biomolécules dans un solvant non aqueux et à coupler le complexe de biomolécules à un substrat en présence du solvant non aqueux. Le substrat ou le support solide est une particule de polymère, la biomolécule à immobiliser sur la particule est un oligonucléotide, le contre-ion lipophile est un ion de tétrabutylammonium et le solvant non aqueux est un N-méthylpyrrolidone (NMP). Les particules enrobées d'oligonucléotide sont utilisées dans l'isolement d'acides nucléiques ou dans des réactions de séquençage d'acide nucléique.
