Abstract:
(EN) Methods and compositions are provided to measure the binding of a test compound to a target peptide by measuring the effect of the compound on the abundance of the target peptide inside a cell. The target peptide may bind the test compound at an active site or an allosteric site, and it has been found that such binding may stabilize the target peptide against cellular degradation. The target peptide will preferably comprise a destabilizing mutation which shortens the half life of the target peptide within the cell, typically a mammalian cell. Test compounds, including small molecules, have been found to stabilize target peptides. Also provided are systems and kits for use in practicing the methods.
(FR) Procédés et compositions permettant de mesurer la liaison d'un composé d'essai vers un peptide cible en mesurant l'effet du composé sur l'abondance du peptide cible à l'intérieur d'une cellule. Le peptide cible peut lier le composé d'essai au niveau d'un site actif ou un site allostérique, et il a été découvert qu'une telle liaison peut stabiliser le peptide cible quant à la dégradation cellulaire. Le peptide cible comprend de préférence une mutation de déstabilisation qui raccourcit la demi-vie du peptide cible à l'intérieur de la cellule, typiquement une cellule de mammifère. Des composés d'essai, comprenant de petites molécules, ont été trouvés pour stabiliser des peptides cibles. L'invention porte également sur des systèmes et des kits d'utilisation pour la mise en pratique des procédés.
