Abstract:
(EN) The present invention relates to an apparatus and method for automatically analyzing biological samples, which: dissolve a biological sample in a hydrolase and cell lysate, and dissolve nucleic acid in a solvent, and then attach same to magnetic particles and wash same; and then ultimately wash same using an organic solvent and dry the magnetic particles using a vacuum pump; and then liquate a target nucleic acid attached to the magnetic particles in a solvent; and then apply same to a vessel including a nucleic acid-based amplification reagent and mix same; and then adjust the temperature for amplification and simultaneously irradiate excitation light at a reactor and detect amplification in real time by measuring fluorescent light, deactivate the amplified product by using an infrared lamp after the amplification, and then acquire an image through electrophoresis; and use one apparatus for the entire process of analyzing molecular mass. Thus, after mounting biological sample kits, an electronically automated process can be implemented in order to perform a qualitative and quantitative inspection having a high level of accuracy and reproducibility. In particular, an infrared lamp is provided having a movable function in order to concentrate radiation on an amplified product and to deactivate a genetically amplified product. Thus, false-positives can fundamentally be prevented.
(FR) La présente invention concerne un appareil et un procédé d'analyse automatique d'échantillons biologiques, le procédé comprenant : la dissolution d'un échantillon biologique dans une hydrolase et un lysat cellulaire, la dissolution d'un acide nucléique dans un solvant, la fixation de l'acide nucléique sur des particules magnétiques et le lavage des particules magnétiques ; le lavage des particules magnétiques avec un solvant organique et leur séchage sous vide ; l'agrégation d'un acide nucléique cible fixé aux particules magnétiques dans un solvant ; l'introduction des particules magnétiques dans un récipient contenant un réactif d'amplification à base d'acides nucléiques et leur mélange ; l'ajustement de la température pour effectuer une amplification, l'irradiation simultanée d'une lumière d'excitation dans un réacteur, la détection en temps réel d'une amplification par la mesure d'une lumière de fluorescence, la désactivation du produit amplifié au moyen d'une lampe infrarouge après l'amplification et l'acquisition d'une image par électrophorèse ; et l'utilisation d'un appareil pour la totalité du processus d'analyse de masse moléculaire. Ainsi, après le montage de kits d'échantillons biologiques, un procédé automatisé électroniquement peut être mis en œuvre afin d'effectuer une inspection qualitative et quantitative avec un niveau élevé de précision et de reproductibilité. En particulier, il est utilisé une lampe infrarouge possédant une fonction mobile afin de concentrer le rayonnement sur un produit amplifié et de désactiver un produit génétiquement amplifié. Ceci permet d'éviter les faux positifs.
(KO) 본 발명은 생물학적 시료를 단백질가수분해 효소 및 세포용해액에 용해시켜 핵산을 용액에 녹인 후 자성입자에 부착시켜 세척한 후 최종적으로 유기용매로 세척하고 자성입자를 진공펌프로 건조한 다음 자성입자에 부착된 타겟핵산을 수용액에 용출한 후, 핵산증폭시약을 포함하고 있는 용기에 가해주고 혼합한 후 온도를 조절하여 증폭하면서 동시에 여기광을 반응기에 조사하여 형광을 측정함으로서 실시간으로 증폭을 검출하고, 증폭 후에 자외선램프로 증폭산물을 불활화시킨 후 전기영동을 통해 영상을 획득하고 분자량을 분석하는 전 과정을 하나의 장치를 이용할 수 있으며 생물학적 시료와 관련 키트들을 장착한 후 전자동으로 수행하여 정확도와 재현성 높은 있는 정성, 정량검사를 수행할 수 있고, 특히 자외선램프가 이동가능하게 구비되어 증폭산물을 집중적으로 조사하여 유전자증폭산물을 불활화시킬 수 있고, 위양성을 근본적으로 막을 수 있는 생물학적 시료 자동 분석 장치 및 방법에 관한 것이다.
