Abstract:
(EN) The invention relates to a method for linking at least two target nucleic acid molecules from a single biological compartment, comprising the steps of isolating a fraction from a sample, wherein the fraction comprises the compartment comprising at least two nucleic acid molecules; diluting said fraction and aliquoting the dilution in multiple separate reaction vessels such that each reaction vessel comprises preferably one compartment, or encapsulating said compartment in emulsion droplets such that each droplet comprises preferably one compartment; linking said at least two target nucleic acid molecules, preferably by overlap extension PCR. The method may be employed in the analysis of mutations present in a single cell and in the production of antibodies which are present in a single hybridoma.
(FR) La présente invention concerne un procédé de liaison d'au moins deux molécules d'acides nucléiques cibles à partir d'un seul compartiment biologique, ledit procédé comprenant les étapes consistant à isoler une fraction à partir d'un échantillon, la fraction comprenant le compartiment comprenant au moins deux molécules d'acides nucléiques; diluer ladite fraction et placer des parties aliquotes de la dilution dans de multiples récipients de réaction séparés de sorte que chaque récipient de réaction comprenne de préférence un compartiment, ou encapsuler ledit compartiment dans des gouttelettes d'émulsion de sorte que chaque gouttelette comprenne de préférence un compartiment; lier lesdites au moins deux molécules d'acides nucléiques cibles, de préférence par PCR par chevauchement-extension. Le procédé peut être utilisé dans l'analyse de mutations présentes dans une seule cellule et dans la production d'anticorps qui sont présents dans un seul hybridome.
