(EN)Synthetic methylated peptides derived from the tau protein of paired helical filaments and non-diseased control brain are disclosed. Alzheimer's disease specificity is provided by the presence or absence of methyl moieties on lysine residues and differences between mono-, di-, and tri-methylation. The methylated peptide is useful as an antigen and a binding partner for identifying compounds that interact with the peptide and the methylated tau protein, including antibodies that can distinguish non-diseased brain from that affected by Alzheimer's disease. The resulting antibodies are useful diagnostically and therapeutically. The compounds that specifically bind to methylated tau proteins are useful for eliminating abnormally methylated tau.
(FR)L'invention concerne des peptides méthylés synthétiques issus de la protéine tau de filaments hélicoïdaux appariés et d'un cerveau témoin non malade. La spécificité de la maladie d'Alzheimer est apportée par la présence ou l'absence de fractions méthyles sur des résidus lysine et des différences entre une mono-, une di- et une tri-méthylation. Le peptide méthylé est utile en tant qu'antigène et partenaire de liaison pour l'identification de composés qui interagissent avec le peptide et la protéine tau méthylée, comprenant des anticorps qui peuvent distinguer un cerveau non malade d'un cerveau affecté par la maladie d'Alzheimer.
