(EN)The invention relates to a molecular biology method and kit that associate human proteins XRCC4 and XLF with a ligase of microbial origin, such as ligase T4, which can improve in-vitro DNA molecule ligation methods. More specifically, the method allows the efficiency of intermolecular ligation reactions to be improved, in particular during operations involving the cloning or assembling of DNA fragments in order to form DNA vectors or constructs.
(FR)L'invention se rapporte à un procédé et à un kit de biologie moléculaire associant les protéines XRCC4 et XLF humaines et une ligase d'origine microbienne, telle que la ligase T4, permettant d'améliorer les procédés in-vitro de ligation de molécules d'ADN. Ce procédé permet plus particulièrement d'améliorer le rendement des réactions de ligations intermoléculaires, notamment, lors de manipulation de clonage ou d'assemblage de fragments d'ADN pour former des vecteurs ou des constructions d'ADN.
