Abstract:
(EN) The present invention relates to a microfluidic device for extracting and isolating DNA from cells. The device includes a support having an inlet port for receiving a sample containing a cell, an outlet port for dispensing DNA isolated from the cell, and a microfluidic channel disposed within the support and extending from the inlet port to the outlet port. The microfluidic channel includes a micropillar array, an inflow channel disposed between the inlet port and the micropillar array, and an outflow channel disposed between the micropillar array and the outlet port. The micropillar array includes micropillars spatially configured to entrap, by size exclusion, the cell, to immobilize DNA released from the cell, and to maintain the immobilized DNA in elongated or non-elongated form when hydrodynamic force is applied to the microfluidic channel. Systems and methods of making and using the device are also provided herein.
(FR) La présente invention concerne un dispositif microfluidique pour l'extraction et l'isolement d'ADN à partir de cellules. Le dispositif comprend un support ayant un orifice d'entrée pour recevoir un échantillon contenant une cellule, un orifice de sortie pour distribuer l'ADN isolé à partir de la cellule et un canal microfluidique disposé à l'intérieur du support et s'étendant de l'orifice d'entrée à l'orifice de sortie. Le canal microfluidique comprend un réseau à micropiliers, un canal d'écoulement entrant disposé entre l'orifice d'entrée et le réseau à micropiliers et un canal d'écoulement sortant disposé entre le réseau à micropiliers et l'orifice de sortie. Le réseau à micropiliers comprend des micropiliers configurés spatialement pour capturer, par exclusion stérique, la cellule, pour immobiliser l'ADN libéré de la cellule et pour maintenir l'ADN immobilisé sous une forme allongée ou non allongée lorsqu'une force hydrodynamique est appliquée au canal microfluidique. La présente invention concerne également des systèmes et procédés de fabrication et d'utilisation du dispositif.
