(EN)The present invention relates to a transgenic pig secreting sTNFR1-Fc, which has inserted therein genes coding for sTNFR1-Fc having an extracellular region for the soluble tumor necrosis factor receptor (sTNFR1) which is fused to an Fc region of an immunoglobulin. The present invention also relates to a method for producing the transgenic pig, organs separated from the transgenic pig, a somatic cell donor cell line in which sTNFR1-Fc genes are inserted, a method for preparing blood including sTNFR1-Fc and to a method for preparing human sTNFR1-Fc from the blood of the transgenic pig. The transgenic pig may secrete substances for inhibiting TNF-α activity in blood, and thus may inhibit immune response and inflammation response, and therefore may be valuably used in heterotransplantation. In addition, the transgenic pig of the present invention has type O blood, and therefore may be transplanted for inhibiting an inflammatory response regardless of blood type.
(FR)Cette invention concerne un porc transgénique sécrétant des sTNFR1-Fc, chez lequel des gènes codant pour sTNFR1-Fc ont été insérés, lesdits sTNFR1-Fc comprenant une région extracellulaire pour le récepteur du facteur de croissance tumorale soluble (sTNFR1) qui est liée par fusion à la région Fc d'une immunoglobuline. Cette invention concerne également un procédé de production dudit porc transgénique, d'organes séparés dudit porc transgénique, d'une lignée cellulaire donneur de cellules somatiques dans laquelle des gènes sTNFR1-Fc sont insérés, un procédé de préparation de sang contenant des sTNFR1-Fc et un procédé de préparation de sTNFR1-Fc humains à partir du sang du porc transgénique. Le porc transgénique selon l'invention peut sécréter des substances capables d'inhiber l'activité TNF-α dans le sang, et par conséquent, capables d'inhiber la réponse immunitaire et la réponse inflammatoire, ce qui permet de les utiliser de manière avantageuse dans le domaine des hétérogreffes. De plus, le porc transgénique selon l'invention ayant un sang du groupe O, il peut par conséquent être utilisé dans une greffe pour inhiber une réponse inflammatoire, indépendamment du groupe sanguin.
(KO)본 발명은 인간 sTNFR1(soluble tumor necrosis factor receptor)의 세포 외역 부위와 면역글로불린의 Fc 영역이 융합된 sTNFR1-Fc를 코딩하는 유전자가 삽입된, sTNFR1-Fc를 분비하는 형질전환 돼지, 이의 제조방법, 상기 형질전환 돼지로부터 분리된 장기, sTNFR1-Fc 유전자가 삽입된 체세포 공여 세포주, sTNFR1-Fc를 포함하는 혈액 제조방법, 및 형질전환 돼지의 혈액으로부터 인간 sTNFR1-Fc의 제조방법에 관한 것이다. 형질전환 돼지는 혈액 내에서 TNF-α의 활성을 억제시키는 물질을 분비함으로써, 면역 반응 및 염증 반응을 억제시킬 수 있어, 이종간 이식에 유용하게 사용될 수 있다. 또한 O형 돼지로서 혈액 타입에 상관없이 염증반응 억제를 위해 이식될 수 있다.
