(EN)The present invention relates to a method for determining the single nucleotide polymorphism of target genes using a real-time polymerase chain reaction, comprising the following steps: filling different tubes with allele-specific primers of target genes and carrying out real-time polymerase chain reaction using each probe common to target genes to obtain amplification products of the target genes; obtaining a Ct value from the allele-specific amplification products of each tube; obtaining a ΔCt value between alleles from the Ct value obtained in the previous step; and determining the single nucleotide polymorphism of the target genes from the obtained ΔCt value. The method for determining single nucleotide polymorphism according to the present invention may advantageously be used to detect a desired SNP in a short time.
(FR)Cette invention concerne un procédé permettant de déterminer le polymorphisme d'un seul nucléotide de gènes cibles faisant appel à une réaction en chaîne de la polymérase en temps réel, ledit procédé comprenant les étapes suivantes : remplir différents tubes avec des amorces spécifiques d'allèles des gènes cibles et procéder à une réaction en chaîne de la polymérase en temps réel à l'aide de chaque sonde commune aux gènes cibles pour obtenir des produits d'amplification des gènes cibles ; obtenir une valeur Ct à partir des produits d'amplification spécifiques d'allèles de chaque tube ; obtenir une valeur ΔCt entre les allèles à partir de la valeur Ct obtenue dans l'étape précédente ; et déterminer le polymorphisme d'un seul nucléotide des gènes cibles à partir de la valeur ΔCt obtenue. Le procédé permettant de déterminer le polymorphisme d'un seul nucléotide selon la présente invention peut être utilisé de manière avantageuse pour détecter un SNP souhaité en un court laps de temps.
(KO)본 발명은 표적 유전자의 대립형질 특이적 프라이머를 서로 다른 튜브에 포함하고 표적 유전자에 공통적인 프로브를 각각 사용하여 실시간 중합효소 연쇄반응을 수행하여 상기 표적 유전자의 증폭산물을 생성하는 단계; 상기 각각의 튜브의 대립형질 특이적 증폭산물로부터 Ct 값을 구하는 단계; 상기 단계 (2)에서 얻어진 Ct 값으로부터 대립형질간의 ΔCt 값을 구하는 단계; 및 상기 얻어진 ΔCt 값으로부터 상기 표적 유전자의 단일 염기 다형성을 판별하는 단계를 포함하는 실시간 중합효소 연쇄반응을 이용한 표적 유전자의 단일 염기 다형성 판별 방법에 관한 것이다. 본 발명의 표적 유전자의 단일 염기 다형성 판별 방법은 빠른 시간 내에 원하는 SNP를 검출할 수 있다는 장점이 있다.
