(EN)The effects on human hair growth of 7'-phloroeckol (1-(3',5'-dihydroxyphenoxy)-7-(2",4",6-trihydroxyphenoxy)-2,4,9-trihydroxy­dibenzo-1,4-dioxin) isolated from E. cava (Ecklonia cava) have been studied in cell systems. The cell-proliferation and anti-apoptotic effects on human human hair follicle dermal papilla cells (HFDPC) and human keratinocytes (human epidermal keratinocytes: HaCaT) were investigated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay and western blot. The 7'-phloroeckol significantly increased HFDPC and HaCaT proliferation. With HFDPC and HaCaT, the amount of PI3K and phosphorylated AKT protein expression increased in a way which was dependent on the extent of 7'-phloroeckol treatment. Also, the levels of MEK1/2, phosphorylated ERK and c-Fos proteins, which are associated with cell proliferation, increased in a way which was dependent on the extent of 7'-phloroeckol treatment. Meanwhile, apoptosis was inhibited as there was an increase in the Bcl-2/Bax ratio which is associated with apoptosis, and there was a significant reduction in p53 and also caspase-3 and caspase-9. These results indicate that 7'-phloroeckol can promote human hair growth by inhibiting apoptosis and stimulating HFDPC and HaCaT proliferation.
(FR)Selon l'invention, les effets sur la croissance des cheveux humains du 7'-phloroeckol (1-(3'-5'-dihydroxyphénoxy)-7-(2'',4'',6-trihydroxyphénoxy)-2,4,9-trihydroxydibenzo-1,4-dioxine) isolé à partir de E. cava (Ecklonia cava) ont été étudiés dans des systèmes cellulaires. La prolifération cellulaire et les effets anti-apoptotiques sur les cellules de papille dermique du follicule pileux humain (HFDPC) et sur les kératinocytes humains (les kératinocytes épidermiques humains : HaCaT) ont été étudiés à l'aide de l'essai au bromure de 3-(4,5-diméthylthiazol-2-yl)-2,5-diphényltétrazolium (MTT) et par transfert de type Western. Le 7'-phloroeckol a considérablement augmenté la prolifération de HFDPC et de HaCaT. Avec HFDPC et HaCaT, le niveau d'expression des protéines PI3K et AKT phosphorylée a augmenté d'une manière qui était dépendante du niveau de traitement par le 7'-phloroeckol. De plus, les niveaux des protéines MEK1/2, ERK phosphorylée et c-Fos, qui sont associées à la prolifération cellulaire, ont augmenté d'une manière qui était dépendante du niveau de traitement par le 7'-phloroeckol. Simultanément, l'apoptose a été inhibée puisqu'il y a eu une augmentation du rapport Bcl-2/Bax qui est associé à l'apoptose et il y a eu une réduction considérable de p53 et également de la caspase-3 et de la caspase-9. Ces résultats indiquent que le 7'-phloroeckol peut favoriser la croissance de cheveux humains par l'inhibition de l'apoptose et la stimulation de la prolifération de HFDPC et HaCaT.

Cava)로부터 분리한 7'-플로로에콜 (1-(3',5'-다이하이드록시페녹시)-7-(2",4",6-트리하이드록시페녹시)-2,4,9-트리하이드록시다이벤조-1,4-다이옥신)이 인체 모발 성장에 대해 갖는 효과에 대하여 세포 시스템내에서 연구되었다. 세포증식 및 항세포사멸사의 효과를 3-(4,5-다이메틸티아졸-2-일)-2,5-다이페닐 테트라졸륨 브로마이드(MTT) 분석 및 웨스턴 블롯(western blot)에 의해 인체 모낭 모유두 세포(hair follicle dermal papilla cell: HFDPC) 및 인체 각질형성세포(human epidermal keratinocyte: HaCaT)에 대하여 실시하였다. 7'-플로로에콜은 HFDPC 및 HaCaT의 증식을 유의하게 증식시켰다. HFDPC와 HaCaT에서 PI3K 및 인산화-AKT 단백질 발현이 7'-플로로에콜 처리량에 의존적으로 증가하였다. 또한, 세포 증식에 관련된 MEK1/2, 인산화-ERK 및 c-Fos의 단백질 수준은 7'-플로로에콜 처리량 의존적으로 증가하였다. 한편, 세포자멸사에 관련된 Bcl-2/Bax 비가 증가하여 세포자멸사를 저해하였고, p53 및 카스파아제(caspase)-3, 카스파아제-9은 유의하게 감소하였다. 이들 결과는 7'-플로로에콜이 세포자멸사를 저해하고 HFDPC 및 HaCaT의 증식을 자극시킴으로써 인체 모발 성장을 촉진할 수 있음을 시사한다.
