Abstract

A pH responsive and mucoadhesive pharmaceutical dosage form for the release of a pharmaceutically active agent is described. The dosage form includes a mucoadhesive layer for site-specific mucoadhesion, a water-insoluble outer layer, and an intermediate layer including one or more pharmaceutically active agents for site-specific delivery. The different membranous layers perform different functions in order to create a drug delivery system which is able to deliver a drug to a specific site, for a particular period of time and with a specific drug release pattern. The dosage form can have two or more intermediate layers, each layer comprising an active agent. The mucoadhesive layer can also include an active agents. The dosage form is preferably an oral or buccal delivery form for release of the active agent into the gastro intestinal tract. The intermediate layer can be an electrospun fibrous membrane layer containing the active agent.

La présente invention concerne une forme pharmaceutique sensible au pH et mucoadhésive pour la libération d'un agent pharmaceutiquement actif. La forme pharmaceutique comprend une couche mucoadhésive pour mucoadhésion site-spécifique, une couche externe insoluble dans l'eau, et une couche intermédiaire comprenant un ou plusieurs agents pharmaceutiquement actifs pour administration site-spécifique. Les différentes couches membranaires remplissent différentes fonctions afin de créer un système d'administration de médicament qui est en mesure de libérer un médicament à un site spécifique, pendant une durée particulière et avec un profil de libération de médicament spécifique. La forme pharmaceutique peut avoir deux couches intermédiaires ou plus, chaque couche comprenant un agent actif. La couche mucoadhésive peut comprendre en outre un agent actif. La forme pharmaceutique est de préférence une forme pour administration orale ou buccale pour libération de l'agent actif dans le tractus gastro-intestinal. La couche intermédiaire peut être une couche de membrane fibreuse électrofilée contenant l'agent actif.
