(EN)The present invention relates to a multiplex microfluidic device for selecting nucleic acid aptamers, and a high throughput selection method for nucleic acid aptamers using the same, and more specifically, to a multiplex microfluidic device (SELEX lap-on-a-chip) in which a conventional aptamer development method for a single target is converted into an improved multiplex platform, and a high throughput selection method for nucleic acid aptamers in which the method is linked with high throughput sequencing. According to the present invention, the multiplex microfluidic device can simultaneously detect each aptamer for a plurality of targets and remarkably reduces the processing amount and processing time compared with a conventional multiplex method. In particular, the target coupling/separation/amplification process round can be greatly reduced and an automated process can carry out a selection process of aptamers by linking the device of the present invention with high throughput sequencing, and thus the device is very useful.
(FR)La présente invention concerne un dispositif microfluidique multiplexé pour sélectionner des aptamères d'acide nucléique, et un procédé de sélection à haut débit pour aptamères d'acide nucléique utilisant ledit dispositif. L'invention concerne plus spécifiquement un dispositif microfluidique multiplexé (laboratoire sur puce SELEX) dans lequel un procédé de développement d'aptamères conventionnel pour une seule cible est transformé en une plate-forme multiplexée améliorée, et un procédé de sélection à haut débit pour aptamères d'acide nucléique qui est associé à un séquençage à haut débit. Selon la présente invention, le dispositif microfluidique multiplexé peut détecter simultanément chaque aptamère pour une pluralité de cibles et réduit de façon remarquable la quantité et le temps de traitement par rapport à un procédé multiplexé conventionnel. En particulier, la routine de procédés de couplage/séparation/amplification de la cible peut être fortement réduite et un procédé automatisé de sélection d'aptamères peut être mis en œuvre en associant le dispositif de la présente invention à un séquençage à haut débit, le dispositif se révélant ainsi très utile.
(KO)본 발명은 핵산 압타머 선별을 위한 멀티플렉스 미세 유체 장치 및 이를 이용한 핵산 압타머의 고속 선별방법에 관한 것으로, 보다 구체적으로는 기존 단일 타겟에 대한 압타머 개발방식을 개선된 멀티플렉스 플랫폼으로 전환시킨 멀티플렉스 마이크로 유체 장치 (SELEX lap-on-a-chip) 및 이를 고속 시퀀싱법(high throughput sequencing)과 연계한 것을 특징으로 하는 핵산 압타머의 고속 선별방법(high throughput selection mehtod)에 관한 것이다. 본 발명에 따른 멀티플렉스 유체 장치는 다수개의 타겟에 대한 각 압타머를 동시에 검출할 수 있을 뿐만 아니라, 종래의 멀티플렉스 기법에 비하여 처리량 및 처리시간을 크게 감축하는 장점이 있으며, 특히 본 발명에 따른 장치를 고속 시퀀싱법과 연계하여 압타머 선별공정을 수행함으로써 타겟결합/분리/증폭 공정 라운드를 대폭 감소시키고 자동화 공정의 수행을 가능하게 하는바, 매우 유용하다.
