Abstract

Disclosed are: an induced hepatic stem cell useful for safety tests, toxicity tests, metabolism tests, drug interaction tests, anti-viral activity tests, screening tests for medicinal agents such as hyperlipemia therapeutic agents, hypertension therapeutic agents, pharmaceutical low-molecular-weight compound agents and pharmaceutical antibody agents, the screening for potential drug targets, the production of animal models, the production of hepatocyte-produced proteins, and regenerative medicine; a process for producing the cell; and applications of the cell. The induced hepatic stem cell is characterized by fulfilling at least the following requirements (1) to (3): (1) at least 15 genes selected from specific genes that are marker genes for embryonic stem cells are expressed in the cell; (2) the cell has properties of hepatocytes; and (3) the cell can be proliferatively cultured or sub-cultured for at least three days.

La présente invention concerne : une cellule souche hépatique induite utile pour des essais de sécurité, des essais de toxicité, des essais de métabolisme, des essais d'interaction médicamenteuse, des essais d'activité antivirale, des essais de criblage pour des agents médicinaux tels que des agents thérapeutiques contre l'hyperlipidémie, des agents thérapeutiques contre l'hypertension, des agents pharmaceutiques de composé de faible poids moléculaire et des agents pharmaceutiques d'anticorps, le criblage de cibles médicamenteuses potentielles, la production de modèles animaux, la production de protéines produites par des hépatocytes, et la médecine régénérative ; un procédé pour produire la cellule ; et des applications de la cellule. La cellule souche hépatique induite est caractérisée en ce qu'elle satisfait au moins aux exigences suivantes (1) à (3) : (1) au moins 15 gènes choisis parmi des gènes spécifiques qui sont des gènes marqueurs pour des cellules souches embryonnaires sont exprimés dans la cellule ; (2) la cellule a des propriétés d'hépatocytes ; et (3) la cellule peut être cultivée de façon proliférative ou sous-cultivée pendant au moins trois jours.

 本発明は、安全性試験、毒性試験、代謝試験、薬物相互作用試験、抗ウイルス活性試験、高脂血症薬、高血圧治療薬、低分子化合物医薬、抗体医薬などの医薬品のスクリーニング試験、創薬標的スクリーニング、動物モデル作製、肝細胞産生タンパク質の生産、及び再生医療に有用な、下記の誘導肝幹細胞、その製造方法及び該細胞の応用である。 少なくとも下記(1)~(3)の要件を具備することを特徴とする誘導肝幹細胞; (1)胚性幹細胞のマーカー遺伝子である特定の遺伝子群の中から選択された少なくとも15種の遺伝子を発現している (2)肝細胞としての性質を有する (3)3日以上増殖培養又は継代培養可能である。
