The invention relates to a library of 298 peptides useful for formulating a novel therapeutic HIV vaccine. The peptide sequences were selected on the basis of calibration of molecular structure properties of HIV-I or host cell proteins. A mixture of D- and L-amino acids or all D-amino acids are used to synthesize the stereoisomer peptides for the purpose of increasing their stability. The peptides are expected to have the ability to potently inhibit functioning of proteins important for HIV infection. A plural of the peptides are conjugated together with a biocompatible polymer, preferably HPMA to further increase stability and solubility, decrease drug toxicity, and potentially evade multidrug resistance and exert cooperative effect, since some peptides are the ligands for host proteins like integrin, and others which can bring the therapeutic peptides to the target cells and therefore help disrupt the interactions between the host proteins and the HIV proteins.

La présente invention concerne une bibliothèque de 298 peptides utiles dans la formulation d'un nouveau vaccin thérapeutique contre le VIH. Les séquences peptidiques ont été sélectionnées sur la base de la calibration de propriétés de structure moléculaire du VIH-I ou de protéines de cellule hôte. Un mélange d'acides aminés D et d'acides aminés L ou de tous les acides aminés D est utilisé pour synthétiser les peptides stéréo-isomères en vue d'augmenter leur stabilité. Lesdits peptides sont supposés présenter la capacité d'inhiber puissamment le fonctionnement de protéines jouant un rôle important dans l'infection par le VIH. Une pluralité desdits peptides est conjuguée à un polymère biocompatible, de préférence HPMA, en vue d'accroître la stabilité et la solubilité, de diminuer la toxicité du médicament, et potentiellement d'éviter la résistance aux médicaments et d'exercer un effet coopératif ; certains peptides sont en effet des ligands pour protéines hôtes, telles que l'intégrine, et d'autres sont capables d'amener les peptides thérapeutiques aux cellules cibles, et de contribuer ainsi à l'interruption des interactions entre les protéines hôtes et les protéines du VIH.
