(EN)The present invention provides an improved method for the purification of a mixture of complexes comprising a stress protein complexed to a peptide or peptide fragment from a source mixture, typically a cell lysate. The method of the invention provides for protein complexes to be purified using ion exchange chromatography based methods, wherein a modified buffer solution is used which results in the purified stress protein complexes being more immunogenic than protein complexes obtained using conventional methodology. The purified complexes can be used to produce improved vaccine preparations which elicit enhanced immune responses in the subjects to whom the vaccine compositions are administered.
(FR)L'invention porte sur un procédé amélioré de purification d'un mélange de complexes comprenant une protéine de stress complexée à un peptide ou à un fragment de peptide, provenant d'un mélange source, généralement un lysat de cellules. Ledit procédé de purification de complexes de protéines, qui fait appel à des techniques de chromatographie d'échange d'ions faisant intervenir une solution tampon modifiée, permet d'obtenir des complexes de protéines de stress plus immunogènes que ceux obtenus par les techniques usuelles. Ces complexes purifiés peuvent servir à produire des préparations de vaccins améliorées qui provoquent des réponses immunitaires accrues chez les patients à qui ces compositions de vaccins sont administrées.
