(EN)The invention provides a method of producing oligodendrocytes by in vitro differentiation of human multi-potent progenitor cells (MLPCs). The method comprises culturing culturing isolated MLPCs on a first surface in a serum-free defined culture medium; replacing the culture medium with serum-free culture medium supplemented with bFGF, EGF and PDGF-AA for approximately 24 hours; changing the cultured MLPCs into the supplemented serum-free culture medium further supplemented with differentiation factors norepinephrine, forskolin. and K252a; establishing a 3D environment by covering the culture with a second surface opposite and spaced apart from the first surface, so as to contain the MLPCs therebetween; and continuing to culture until a majority of the MLPCs have differentiated into oligodendrocytes. Additionally included is a method of treatment for a subject afflicted by a disease characterized by central or peripheral nervous system demyelination, the method comprising transplanting into the subject oligodendrocytes produced according to the method disclosed.
(FR)L'invention porte sur un procédé de production d'oligodendrocytes par différenciation in vitro de cellules progénitrices multipotentes humaines (MLPC). Le procédé comprend la culture des MLPC isolées en culture sur une première surface dans un milieu de culture défini exempt de sérum; le remplacement du milieu de culture par un milieu de culture exempt de sérum enrichi de bFGF, EGF et PDGF-AA pendant approximativement 24 heures; le changement des MLPC en culture dans le milieu de culture exempt de sérum enrichi, enrichi en outre des facteurs de différenciation norépinéphrine, forscoline et K252a; l'établissement d'un environnement 3D par recouvrement de la culture avec une seconde surface opposée et espacée de la première surface, de façon à contenir les MLPC entre celles-ci; et la poursuite de la culture jusqu'à ce qu'une majorité des MLPC se soient différenciés et produisent des oligodendrocytes.
