Abstract:
(EN) Disclosed are methods for suppressing the innate immune response of a cell to transfection with an exogenous nucleic acid. Further disclosed are methods for increasing expression of a protein encoded by an exogenous nucleic acid by repeated delivery of the exogenous nucleic acid to a cell, and methods of changing the phenotype of a cell by differentiating, transdifferentiating or dedifferentiating cells by repeatedly delivering one or more nucleic acids that encode defined proteins. A method is provided for extended transient transfection by repeated delivery of an in vitro-transcribed RNA ("ivT-RNA") to a cell to achieve a high and sustained level of expression of a protein encoded by an ivT-RNA transcripts.
(FR) L'invention porte sur des procédés de suppression de la réponse immunitaire innée d'une cellule à une transfection par un acide nucléique exogène. L'invention porte en outre sur des procédés pour augmenter l'expression d'une protéine codée par un acide nucléique exogène par une administration répétée dudit acide nucléique exogène à une cellule, et sur des procédés pour changer le phénotype d'une cellule par différentiation, transdifférenciation ou dédifférenciation de cellules par une administration répétée d'un ou plusieurs acides nucléiques qui codent pour des protéines définies. L'invention porte également sur un procédé de transfection transitoire étendue par une administration répétée d'un ARN transcrit in vitro (« ARN ivT ») à une cellule pour permettre un niveau élevé et soutenu d'expression d'une protéine codée par un produit de transcription d'ARN ivT.
