Abstract:
(EN) The present invention relates to a method for in vitro diagnostics of invasive aspergillosis from biological samples taken from the body of a patient using quantitative PCR, which contains the steps of isolation of DNA from the clinical samples, preamplification of fungal DNA using primers targeting conservative part of fungal ribosomal DNA (rDNA), and quantitative real-time PCR detection of species specific region of the internal transcribed spacer 2 (ITS2) gene of an Aspergillus species. It further relates to oligonucleotides useful in this method.
(FR) La présente invention porte sur un procédé de diagnostic in vitro d'une aspergillose invasive à partir d'échantillons biologiques prélevés du corps d'un patient à l'aide d'une PCR quantitative. Ledit procédé comprend les étapes d'isolement d'ADN à partir des échantillons cliniques, de préamplification d'ADN fongique à l'aide d'amorces ciblant des parties de conservation d'ADN ribosomique fongique (ADNr) et de détection par PCR quantitative en temps réel de régions spécifiques d'espèce du gène de l'espaceur transcrit interne 2 (ITS2) d'une espèce Aspergillus. L'invention porte en outre sur des oligonucléotides utilisés dans ce procédé.
