Abstract:
(EN) MicroRNA (miRNA) profiling of cells showed unique miRNA signatures for each of three Akt isoforms. Among differentially regulated miRNA species, the miR-200 family was downregulated in Akt2-expressing cells. Aktl knockdown inhibited expression of miR-200 and promoted TGFβ-induced epithelial-mesenchymal-transition (EMT) and a stem cell like phenotype. Carcinomas developing in MMTV-cErixZ/Aktl7" mice exhibited increased invasiveness because of EMT induced by miR-200 downregulation. EMT was found to be controlled by miRNA species that are regulated by the balance between Aktl and Akt2, rather than overall Akt levels.
(FR) Le profilage des microARN (miARN) des cellules a montré des signatures uniques des miARN pour chacune des trois isoformes Akt. Parmi les espèces de miARN régulées de manière différentielle, la famille miR-200 est régulée à la baisse dans les celldules exprimant l'Akt2. L'inactivation de Akt1 inhibe l'expression de miR-200 et favorise la transition épithéliale-mésenchymateuse (EMT) induite par TGFβ et un phénotype de type cellule souche. Les carcinomes se développant dans des souris MMTV-cErixZ/Akt17'' présentent un caractère invasif accru à cause de l'EMT induite par la régulation à la baisse des miR-200. On a découvert que l'EMT est contrôlée par des espèces miARN, qui sont régulées par l'équilibre entre Akt1 et Akt2, plutôt que par les taux totaux d'Akt.
