Abstract:
(EN) A system and method for heterologous expression of polyketide biosynthetic pathways from streptomycetes hosts in Escherichia coli for the production and discovery of secondary metabolites. Genomic DNA from Streptomyces rimosus encoding the oxytetracycline biosynthetic pathway is inserted into the genome of the surrogate host Myxococcus xanthus. The M. xanthus transcriptional machinery recognizes and uses the streptomycetes promoter regions to express the biosynthetic enzymes. Co-expression in E. coli of S. rimosus oxytetracycline biosythensis enzymes and M. xanthus σ54, a key piece of the M. xanthus transcriptional machinery, enables E. coli to recognize and use the promoters from the S. rimosus oxytetracycline biosynthetic pathway, facilitating production of oxytetracycline.
(FR) L'invention porte sur un système et un procédé pour l'expression hétérologue de voies biosynthétiques de polycétide à partir d'hôtes Streptomyces dans Escherichia coli, pour la production et la découverte de métabolites secondaires. L'ADN génomique provenant de Streptomyces rimosus, codant pour la voie biosynthétique de l'oxytétracycline, est introduit dans le génome de l'hôte porteur Myxococcus xanthus. La machinerie de transcription de M. xanthus reconnaît et utilise les régions promotrices de Streptomyces pour exprimer les enzymes biosynthétiques. Une co-expression dans E. coli d'enzymes de biosynthèse d'oxytétracycline de S. rimosus et de σ54 de M. xanthus, un morceau clé de la machinerie de transcription de M. xanthus, permet à E. coli de reconnaître et d'utiliser les promoteurs provenant de la voie biosynthétique d'oxytétracycline de S. rimomus, facilitant la production d'oxytétracycline.
