Abstract:
(EN) Auxotrophic Agrobacterium and methods employing auxotrophic Agrobacterium are provided. Auxotrophic Agrobacterium may be used in a variety of methods including biologically containing Agrobacterium comprising a transgene and transforming a plant cell without using an Agrobacterium counter-selective agent. Transforming maize immature embryos using an Agrobacterium auxotrophic for thymidine results in comparable transformation efficiency as transformation achieved using prototrophic Agrobacterium. Methods for producing an Agrobacterium thymidine auxotroph and using the auxotroph in transformation methods are disclosed. Transformed tissues and plants produced using methods of the present invention are also provided.
(FR) La présente invention concerne un Agrobacterium auxotrophe et les méthodes employant ledit Agrobacterium auxotrophe. L'Agrobacterium auxotrophe peut être employé dans diverses méthodes, y compris le confinement biologique d'un Agrobacterium comprenant un transgène et la transformation d'une cellule végétale sans utiliser d'agent contre-sélectif vis-à-vis d'Agrobacterium. La transformation d'embryons immatures de maïs en utilisant un Agrobacterium auxotrophe vis-à-vis de la thymidine permet d'obtenir une efficacité de transformation comparable à celle obtenue avec un Agrobacterium prototrophe. La présente invention concerne les méthodes de production d'un Agrobacterium auxotrophe vis-à-vis de la thymidine et l'utilisation de l'auxotrophe dans des méthodes de transformation.
