Abstract:
(EN) Regulatory elements for controlling expression of transgenes in angiogenic tissue are provided. One embodiment provides an isolated nucleic acid having at least 80%, 85%, 90%, 95%, 97%, 99% or 100% sequence identity to SEQ ID NO: I, or a fragment thereof that causes expression of a transgene in angiogenic tissue. Vectors containing SEQ ID NO: 1 and one or more transgenes are also provided. A preferred vector is an adenovirus vector. The transgene can encode a cytotoxin, pro-apoptotic polypeptide, or a therapeutic polypeptide. SEQ ID NO:1 regulates the expression of the transgene such that the transgene is only expressed in angiogenic tissue including, but not limited to arteries feeding ischemic tissues. Methods of using vectors containing SEQ ID NO: 1 are also provided.
(FR) La présente invention concerne des éléments régulateurs pour le contrôle de l'expression de transgènes dans un tissu angiogène. Dans l'un des modes d'application, la présente invention concerne un acide nucléique isolé d'identité de séquence égale à au moins 80 %, 85 %, 90 %, 95 %, 97 %, 99 % ou 100 % vis-à-vis de la séquence SEQ ID NO: I, où l'un de ses fragments provoquant l'expression d'un transgène dans un tissu angiogène. La présente invention concerne également les vecteurs contenant la SEQ ID NO: 1 et un ou plusieurs transgènes. L'un des vecteurs préférés est un vecteur d'adénovirus. Le transgène peut coder une cytotoxine, un polypeptide pro-apoptotique ou un polypeptide thérapeutique. La SEQ ID NO:1 régule l'expression du transgène, de sorte à ce que le transgène soit uniquement exprimé dans les tissus angiogènes, ce qui inclut sans y être limité les tissus ischémiques irriguant les artères.
