(KO) 바이오리액터 배양기에 의한 조직배양 감자종서의 대량생산 및 씨감자 생산방법
Abstract:
(EN) The present invention relates to a method for producing seed potatoes free of virus through the massive propagation of potato microtubers by growing sterile potato stems in a bio-reactor culture device containing MS liquid culture medium reduced to 1/2 to 1/4 of the amount thereof, increasing the used MS liquid culture medium by 1-2 times, and culturing the potato stem in a dark place. Between 7 and 20 potato microtubers having stems with a length of 3-5 cm produced germ-free by tissue-culture and sprouts grown to 2-5cm are sown per 10 ㎠ on a seedbed filled with bed-soil at a depth of 3-6cm. The small seed potatoes grow under the bed-soil, and are cultivated. The potatoes of 5g or more are used as the seed potatoes for farmhouses, and the potatoes of less than 5g are sown on the seedbed to produce the seed potatoes. The small seed potatoes produced by the present invention are capable of convenient storage, can reduce labor forces since the potatoes do not need to be cut when being sown on the open field, and have the advantage of supplying farmhouses with virus-free seed potatoes.
(FR) L'invention concerne un procédé pour produire des pommes de terre de semence exemptes de virus via la propagation massive de microtubes de pomme de terre, par croissance de tiges de pomme de terre stériles dans un dispositif de culture de bioréacteur contenant un milieu de culture MS liquide dont le volume est réduit de 1/2 à ¼ ; augmentation du milieu de culture MS liquide utilisé d'une à deux fois ; et culture de la tige de pomme de terre dans un endroit sombre. Entre 7 and 20 microtubes de pommes de terre ayant des tiges d'une longueur de 3 à 5 cm sont produits sans germe par culture tissulaire, et des pousses germées de 2 à 5cm sont ensemencées par 10 ㎠ sur un lit de semence rempli d'un lit de terre à une profondeur de 3 à 6cm. Les petites pommes de terre de semence poussent sous le lit de semence et sont cultivées. Les pommes de terre de 5g ou plus sont utilisées comme pommes de terre de semence dans les fermes, et les pommes de terre inférieures à 5g sont ensemencées sur le lit de semence pour produire les pommes de terre de semence. Les petites pommes de terre de semence produites par l'invention peuvent être stockées de manière appropriée, permettent de limiter la main d'œuvre du fait que les pommes de terre n'ont pas besoin d'être coupées lorsqu'on les ensemence en plein champ, et présentent l'avantage de fournir aux fermes des pommes de terre de semence sans virus.
(KO) 본 발명은, 1/2~1/4로 감소된 MS 액체배지가 들어있는 바이오리액터 배양기내에서 무균처리된 감자줄기를 증식시킨 후 1~2배로 증가된 MS 액체배지로 교환하여 어두운 곳에서 배양하여 감자종서(potato microtuber)를 대량으로 증식시켜 바이러스 없는 씨감자를 생산하기 위한 방법이다. 조직배양에 의해 무균 생산된 3~5cm 크기의 감자 줄기 또는 싹이 2∼5mm 정도 자란 감자종서를 3~6cm정도의 상토를 채운 육묘판에 10㎠당 7~20개씩 심어 상토 하부에 소형의 씨감자가 생성되면 수확하여 중량 5g 이상인 것은 농가 재배용 씨감자로 사용하고, 미만인 것은 다시 육묘판에 심어 씨감자를 재차 생산하는 방법에 관한 것이다. 본 발명에 의해 생산된 소형의 씨감자는 보관이 편리하고, 노지에 파종할 때 절단할 필요가 없어 노동력을 획기적으로 절감할 수 있고, 무바이러스 씨감자를 단기간에 농가에 보급할 수 있는 장점이 있다.
