Abstract:
(EN) A method for carrying out nucleic acid amplification, includes providing a reaction chamber (31), accommodating an array (36) of nucleic acid probes (37) at respective locations, for hybridizing to respective target nucleic acids; and introducing a solution (50) into the reaction chamber (31), wherein the solution (50) contains primers, capable of binding to target nucleic acids, nucleotides, nucleic acid extending enzymes and a sample including nucleic acids. The a structure of the nucleic acid probes (37) and of the primers so that a hybridization temperature (TH) of the probes (37) is higher than an annealing temperature (TA) of the primers, whereby hybridization and annealing take place in respective separate temperature ranges (RH, RA).
(FR) L'invention porte sur un procédé pour effectuer l'amplification d'acides nucléiques, comprenant la mise à disposition d'une chambre de réaction (31), contenant un réseau (36) de sondes acides nucléiques (37) à des emplacements respectifs, pour l'hybridation à des acides nucléiques cibles respectifs ; et l'introduction d'une solution (50) dans la chambre de réaction (31), la solution (50) contenant des amorces, aptes à se lier à des acides nucléiques cibles, des nucléotides, des enzymes d'allongement d'acide nucléique et un échantillon comprenant des acides nucléiques. La structure des sondes acides nucléiques (37) et celle des amorces sont telles qu'une température d'hybridation (TH) des sondes (37) est supérieure à la température d'annelage (TA) des amorces, l'hybridation et l'annelage ayant lieu dans des plages de température individuelles respectives (RH, RA).
